For me, New York is very much a city thats best explored by just walking around and seeing what you come across, but the highlights that I love to do whenever I am there are:
Walk across the Brooklyn Bridge
Visit the Tenament Museum down on Delancy
Take the Staten Island Ferry
Museum of Modern Art
Eat at Katz's diner
Drink a beer or two at Heartlands Brewery - I prefer the one at the back of Times Square
Visit Grand Union Station - eat at Michael Jordans steak house
Go to Blue Smoke for some jazz and a bucket of fries over 10 years ago -
Indeed NYC is best explored by walking around the different neighborhoods. Some ideas:
+Walk the Brooklyn Bridge
+Top of the Empire State Building
+Walk through Central Park (always entertainment going on) Horse and buggy ride is overpriced
+Pier 17 for street entertainment, shopping, bars and great views
+Williamsburg Brooklyn (Bedford Ave) for food, bars and artists
+Times Square
+Ride a bike down the West Side bike path for scenic views and excellent parks near Battery Park
+Museum of Modern Art or American Museum of Natural History
+Gray Line Double Decker Bus tours (they may be cheesy, but many are informative and will take you all the way up to Harlem)
+Best view of the city's sklyline. Take a ferry or Path train to Hoboken and visit Pier A for a spectacular view. If you enjoy bars there are about 50 of them along one strip in Hoboken. A W Hotel also just opened on the waterfront also.
+Canal street for flea market type shopping
-Don't waste your money on Italian dinning in Little Italy (Food is overpriced and for tourists)
+Bryant Park for lunch
+St. Patrick's Cathedral and while there Rockefeller Center
+Grand Central Station
+Stroll Soho
+Statue of Liberty (Advised this is an all day affair)
+Columbus Circle
+Washington Square
+Meat Packing District (posh dinning and nightlife) over 10 years ago
